Problem 2. Sequence below is a representative
segment of genomic DNA (coding strand of a gene).
ATGCGTACTCCTTAA
2.1 Fill in the complementary stand of the gene
so it’s double-stranded.
2.2 Translate the gene(assuming that ATG up
front is a start codon). The amino acid sequence of the polypeptide
encoded by this gene is:
2.3 You want to mutagenize your fly stock so
you expose them to EMS. Write out the double- stranded DNA after
one round of replication in the presence of EMS. (You should get
two double-stranded segments.) Assume that every nucleotide that
can be affected is affected by EMS.
2.4 You removed EMS from fly culture. Write out
the double-stranded DNA after second round of replication
without the EMS. You should have four
double-stranded segments.
2.5 Translate the four mutagenized genes from
2.4. Write out amino acid sequence of the
product.

2.6 Compare the protein product of each mutant
gene to the original (wild type) polypeptide from
2.2. For each mutant, indicate what kind of
mutation(s) it has based on sequence: nonsense, missense,
frameshift, silent.
